ETV7 limits the antiviral and antitumor efficacy of CD8+ T cells by diverting their fate toward exhaustion

Jan. 15, 2025

Terminal exhaustion is a critical barrier to antitumor immunity. By integrating and analyzing single-cell RNA-sequencing and single-cell assay for transposase-accessible chromatin with sequencing data, we found that ETS variant 7 (ETV7) is indispensable for determining CD8+ T cell fate in tumors.

Structural basis of human VANGL-PRICKLE interaction

Jan. 09, 2025

Planar cell polarity (PCP) is an evolutionarily conserved process for development and morphogenesis in metazoans. The well-organized polarity pattern in cells is established by the asymmetric distribution of two core protein complexes on opposite sides of the cell membrane.

A rapid chemical reprogramming system to generate human pluripotent stem cells

Jan. 09, 2025

Chemical reprogramming enables the generation of human pluripotent stem (hCiPS) cells from somatic cells using small molecules, providing a promising strategy for regenerative medicine. However, the current method is time consuming, and some cell lines from different donors are resistant to chemical induction, limiting the utility of this approach.

Quantifying the regulatory potential of genetic variants via a hybrid sequence-oriented model with SVEN

Jan. 09, 2025

Deciphering how noncoding DNA determines gene expression is critical for decoding the functional genome. Understanding the transcription effects of noncoding genetic variants are still major unsolved problems, which is critical for downstream applications in human genetics and precision medicine.

Zinc homeostasis regulates caspase activity and inflammasome activation

Dec. 20, 2024

Inflammasome activation drives pyroptotic cell death and the release of inflammatory cytokines, and many diseases involve its overactivation. Zinc is essential for all organisms as a trace element, but its functions in innate immunity remain undefined.
